SEMAGLUTIDE CALCULATOR
Semaglutide is a GLP-1 receptor agonist used for weight loss and metabolic research. Dosing requires slow titration starting at 0.25mg weekly, increasing every 4 weeks to avoid severe nausea and GI side effects.
This calculator shows your exact weekly dose based on which week you're in, calculates injection volume from your concentration, and displays the full titration schedule from 0.25mg up to 2.4mg.
Standard semaglutide titration:
Weeks 1-4: 0.25mg once weekly
Weeks 5-8: 0.5mg once weekly
Weeks 9-12: 1.0mg once weekly
Weeks 13-16: 1.7mg once weekly
Weeks 17+: 2.4mg once weekly (max dose)
Jumping doses too fast = severe nausea, vomiting, GI distress. The slow increase lets your body adapt. Never skip titration steps.
Example: 5mg vial + 2ml water = 2,500mcg/ml concentration
Week 1-4 at 0.25mg (250mcg) = draw 0.10ml
Week 5-8 at 0.5mg (500mcg) = draw 0.20ml
Week 9-12 at 1.0mg (1,000mcg) = draw 0.40ml
HOW
Weekly dosing protocol and titration schedule
Week 1-4: Starting dose (0.25mg weekly)
Purpose: Assess tolerance, minimal side effects
Weight loss: Minimal to none expected
Side effects: Mild nausea possible, usually tolerable
Injection: Once per week, same day each week
Week 5-8: First increase (0.5mg weekly)
Purpose: Begin seeing metabolic effects
Weight loss: 1-2 lbs per week may start
Side effects: Moderate nausea, reduced appetite
Some stay at this dose if effective
Week 9-12: Therapeutic dose (1.0mg weekly)
Purpose: Standard therapeutic level
Weight loss: 2-3 lbs per week typical
Side effects: Nausea manageable for most
Many people's target maintenance dose
Week 13-16: Higher dose (1.7mg weekly)
Purpose: Enhanced weight loss if 1mg insufficient
Weight loss: 2-4 lbs per week
Side effects: Increased GI issues
Not everyone needs to reach this level
Week 17+: Maximum dose (2.4mg weekly)
Purpose: Maximum therapeutic effect
Weight loss: 3-5 lbs per week possible
Side effects: Most intense, GI issues common
Stay here or reduce to tolerable dose
Can you titrate faster?
Not recommended. Some people try 2-week increases instead of 4-week. Higher risk of:
Severe nausea and vomiting
Inability to eat
Dehydration
Abandoning protocol due to side effects
The 4-week steps exist for a reason. GLP-1 side effects are cumulative. Your body needs time to adapt.
Can you stay at lower doses?
Yes. Many people find their sweet spot:
0.5mg weekly: Mild appetite suppression, minimal sides
1.0mg weekly: Good results, tolerable sides (most common maintenance)
1.7mg weekly: Strong effects if you tolerate it
Don't feel obligated to reach 2.4mg. Find the dose that works for YOUR goals and tolerance.
